Study of Anti-Obesity Effect of Rumex Japonicus Houttuyn Ethanol Extracts in 3T3-L1 Preadipocytes cells

Abstract
Rumex japonicus Houttuyn is known for its antiinflammatory and anti-microbial effects. In this study, we determined the effects of ethanol extract of Rumex japonicus Houttuyn (RJHE) on anti-obesity activities in 3T3-L1 preadipocytes. In vitro experiments to prevent obesity and improve anti-obesity effects, research using 3T3-L1 preadipocytes is essential, it also distinctly represents morphologic and molecular changes. Our results showed that RJHE decreased the differentiation of 3T3-L1 preadipocytesusing oil red O staining through decreasing the lipid droplet and had no cytotoxicity in 3T3-L1 preadipocytes using WST-1 assay (100 ¥ìg/mL and 200 ¥ìg/mL). Also, the concentrations of triglycerides(TG) were gradually reduced in 3T3-L1 preadipocytes cells with RJHE (N: 100%, 100 ¥ìg/mL: 64.5%, 200 ¥ìg/mL: 38.1%) using ELISA assay. And RJHE suppressed the Expression of PPAR¥ã, C/EBP¥á and Acetyl-CoA Carboxylase (ACC) in a dose-dependent manner by Western blot analysis. Taken together, our results demonstrate that RJHE indicates anti-obesity effects in vitro situation and has potential uses in chemotherapeutic drugs.
